Please go through the FAQ section of CDER NextGen portal, which can be found in bottom portion of the website, which provides the details that you are looking for.
Since you have access to your old boss email ID, send an Email from that account to CDER Platform Support at EDMSupport@fda.hhs.gov, to inform about the replacement to be made providing reasons and giving details of new replacement persons details including email id.
Read through the manual to get familiar with the CDER NextGen portal.

Hi everyone,
As a new member of RAPS, I wanted to express how much I am enjoying my experience so far.
I have encountered a question for which I have been unable to find an answer. Recently, my boss left the company after establishing a communication thread with FDA through CDER NextGen Portal. As such, we have prepared and uploaded letter to inform FDA of the new contact person. However, since we now have a new contact person with a different email address, I am unsure if we need to create a new account using the new contact email address. Please note that we will still have access to my boss's email account.
